Defoamers Test Method---Fermentation

Defoamer type:  Silicone¨  Polyetherþ  fatty alcohol ¨  Mineral oil ¨ Others ¨

1. Defoamer performance: (Air drum test)defoamers test method

Accurately weigh 200ml of foaming medium with a 1000ml measuring cylinder, turn on the air bubble to the specified foam and mention 800ml and record the blank time, then add the prepared defoamer sample, stir it evenly, then turn on the air bubble and record the volume V changes with time.

2. Solid content test (instrument: oven, Surface dish)

  1. Turn on the oven, set the temperature to 105°C, wash and dry the watch glass
  2. Weigh a certain amount of defoamer sample on the watch glass, and record the weight of the watch glass before and after adding the defoamer as ml and m2
  3. Put the watch glass with the sample in the oven, close the oven after 1h, put the watch glass in the desiccator and cool it, then weigh it as m3;
  4. The solid content of the sample is: W=(m3-m1)/(m2-m1)*100%
  • 3.Viscosity (instrument: mechanical viscometer, constant temperature water bath)

  1. Turn on the water bath and set to the desired temperature
  2. Wash the viscometer, add a standard amount of defoamer sample, turn on the circulating water, and keep the temperature constant for 15min
  3. Calibrate the viscometer, select the appropriate rotor, turn on the viscometer, read and record
  • Particle size: (Test D50, D90 and tail peak with a particle size analyzer)

Turn on the power of the dispersing device of the particle size analyzer and the optical system, warm up for about 30 minutes, and then start the computer. Run the laser particle size analyzer program.

  • Cloud point

  1. If the polyether is soluble in water, use distilled water to prepare a% polyether aqueous solution;
  2. If polyether is insoluble in water, first prepare 10% M224 aqueous solution with distilled water, and then use M224 aqueous solution to prepare% polyether solution.
  Specific steps:

Pour the polyether solution prepared above into a 10ml centrifuge tube and place it in a water bath for heating. While heating the centrifuge tube, stir the polyether solution up and down with a thermometer at the same time. After the solution begins to become cloudy, remove After centrifuging the tube, the temperature is lowered in ice water or tap water. During the cooling process, the polyether solution is continuously stirred up and down, and the temperature at which the polyether solution becomes clear is recorded. This temperature is the cloud point of the polyether.

6. Hydroxyl value (phthalic anhydride titration method)

  • Thermal stability (observing the temperature or precipitation of oil bleaching or precipitation during heating)

Quality of an antifoam is given by two main characteristics : • Knockdown ability or shock effect :Meaning the speed to destroy an existing foam. • Persistence or durability : Meaning the efficiency over time.   These two parameters can be assessed by several tests.
